Two Cancer Therapies Extend Survival in Major Human Trials

Evidence level: Randomized Phase 3 survival outcomes in defined patient populations. Neither therapy is a general anti-aging intervention; both remain investigational for the uses discussed.

1. Divarasib for KRAS G12C-mutated lung cancer

The Krascendo 1 randomized trial studied 338 adults with previously treated advanced non-small-cell lung cancer carrying the KRAS G12C mutation. Investigational divarasib was compared with approved KRAS G12C inhibitors sotorasib or adagrasib.

OutcomeDivarasibComparator
Median overall survival17.7 months12.0 months
Median progression-free survival9.8 months5.7 months
Objective response rate58%32%

Overall survival hazard ratio: 0.56 (95% CI 0.41–0.78). Serious treatment-related adverse events were reported in 7.8% versus 15.2%, respectively. These are group-level outcomes and cannot predict individual survival.

2. Sonesitatug vedotin for CLDN18.2-positive gastrointestinal cancer

The randomized CLARITY-Gastric01 trial enrolled 532 people with previously treated advanced gastric, gastroesophageal-junction or esophageal adenocarcinoma expressing CLDN18.2. Sonesitatug vedotin is an investigational antibody-drug conjugate carrying a cytotoxic payload to CLDN18.2-expressing tumor cells.

OutcomeSonesitatug vedotinInvestigator's choice
Median overall survival, third line or later10.0 months5.0 months
12-month survival, third line or later38.1%18.1%
Median overall survival, full second-line-or-later group10.6 months6.4 months

The third-line-or-later overall-survival hazard ratio was 0.47 (95% CI 0.36–0.62). The trial did not meet its other dual primary endpoint: centrally assessed progression-free survival in the overall population (reported p=0.053). Grade 3 or higher treatment-related adverse events occurred in 36.4% with sonesitatug vedotin and 44.8% with investigator's choice; gastrointestinal side effects remained important. As of October 26, detailed results were available from a conference presentation and sponsor reporting rather than a complete peer-reviewed publication.

Why these studies matter for healthspan

Unlike laboratory biomarkers or animal lifespan results, these trials measure actual survival in people. They may offer eligible patients more time alive following serious illness. They do not show that biological aging slows, healthy people live longer, or immune systems are generally rejuvenated. Trial populations, adverse effects, access and regulatory status all matter.
